Sodium Hyaluronate Usage and Synthesis
Physical and chemical properties:
Sodium hyaluronate, also known as sodium hyaluronate, is a physiologically active substance widely present in the human body. It is a high molecular weight straight-chain mucopolysaccharide composed of disaccharide units composed of glucuronic acid and acetylhexosamine, with a molecular weight of 1 million. It forms a viscous and elastic solution in water with physiological pH and ionic strength. Its molecular morphology is variable, so it can also be passed through with a thinner injection needle. The substance that does not cause inflammation is extracted from sodium hyaluronate and is called Healon. When 10 mg of this product is dissolved in 1 ml of normal saline, its viscosity can be 200,000 times higher than that of aqueous humor or normal saline. The protein content of Healon is less than 0.5%, and it is a sterile high-purity solution.
Pharmacological action:
Sodium hyaluronate promotes wound healing. Sodium hyaluronate can form a macromolecular mesh barrier on the surface of ocular tissue to prevent inflammatory substances from entering the wound and lesion area, and can significantly increase the viscosity and adhesion of the drug solution, so that the drug can be retained on the corneal surface for a long time and in a concentrated manner, achieving a long-lasting and strong effect. Sodium hyaluronate can stabilize the tear film, prolong the tear film rupture time, and relieve dry eye symptoms. It is used for corneal burns caused by keratitis, corneal ulcers, corneal damage or other chemical and physical factors.
Use:
Used for dry eye syndrome, widely used in high-end cosmetics industry.
Production method:
It is a salt formed by the carboxyl group of hyaluronic acid and metallic sodium ions. Hyaluronic acid is widely present in animals and humans, such as skin, muscle, cartilage, brain, cockscomb, artery and vein walls, etc. Commonly used extraction materials include rooster combs, umbilical cords, cow eyes, etc. The preparation method using cockscomb as raw material is as follows.
Extraction: Wash fresh cockscomb with acetone, dehydrate and crush, add Chemicalbook distilled water to soak and extract for 24 hours, repeat 3 times, and combine the filtrate.
Protein removal and precipitation: Mix the extract with an equal volume of chloroform and stir for 3 hours, separate the aqueous phase, add 2 times the amount of ethanol, collect the precipitate, dehydrate with acetone, and vacuum dry to obtain crude hyaluronic acid.
3. Then enzymatic hydrolysis, complexation, dissociation, and precipitation to obtain fine hyaluronic acid. Then salt with metallic sodium ions.
