L-tert-Leucine Usage and Synthesis
Applications
L-tert-Leucine is a key side chain in the synthesis of the antiviral drug atazanavir, which is an antiretroviral drug of the protease inhibitor class that can be used to treat human immunodeficiency virus (HIV) infection.
It can also be used for:
(1) as a key precursor for the synthesis of the chiral phosphino-oxazoline ligand (S)-tert-butylphosphine;
(2) in the synthesis of chiral copper(II) polymers, it can catalyze the kinetic resolution of secondary alcohols by acylation;
(3) in the synthesis of 6-alkyl/acylphenanthridines in metal-free tandem radical cyclization reactions;
(4) in the preparation of tert-Leucine-derived N-acetylthiazolidinethione auxiliary, it provides a high level of diastereoselectivity in the reaction of acetic acid aldehyde with various aldehydes.
Applications
L-tert-Leucine can be used as a catalyst for the enantioselective oxidative coupling and cyclization of hydroquinone compounds to oxa[9]helicene.
Used as a nutritional enhancer, animal feed additive, and for synthetic drugs.
Amino acids are the basic components of proteins, and one of their main physiological functions is to serve as raw materials for protein synthesis. They appear in the body in free or combined states. Proteins in the human body are broken down to produce the following amino acids: alanine, arginine, aspartic acid, asparagine, cysteine, lysine, methionine, phenylalanine, serine, threonine, tryptophan, tyrosine, and valine.
Production method
1. Pivalaldehyde, L-phenylglycinol and trimethylsilyl cyanide are subjected to asymmetric silanization reaction under the action of catalyst A, and then post-treated to obtain compound I;
2. Compound I is hydrolyzed with Chemicalbook under acidic conditions, and post-treated to obtain compound II;
3. Compound II is subjected to catalytic hydrogenation reaction to obtain L-tert-leucine; the catalyst A is magnesium diiodide, magnesium dibromide, magnesium dichloride, magnesium perchlorate or magnesium trifluoromethanesulfonate.
